Overview
Vonaday Tablet is a once-daily fixed-dose antiretroviral combination containing efavirenz 600 mg, lamivudine 300 mg and tenofovir disoproxil fumarate 300 mg. Efavirenz is an NNRTI; lamivudine and tenofovir are NRTIs. The three medicines suppress HIV at complementary points in reverse transcription.
Efavirenz 600 mg-based single-tablet regimens were widely used as first-line ART for many years. Current international guidelines often prefer integrase-inhibitor-based regimens such as dolutegravir because of improved tolerability and resistance profile, but efavirenz 600 mg tablet-based therapy remains an established alternative in selected settings.

Uses and Indications
Vonaday Tablet is prescribed for the treatment of:
- HIV-1 Infection
This fixed-dose combination is used for the treatment of HIV and is not indicated for PrEP due to the efavirenz tablet contained in this drug, which makes a full treatment regimen. The doctor reviews the resistance profile to HIV, renal function, presence of hepatitis B, pregnancy conditions, and other drugs used before taking this combination or when switching to this drug. Tenofovir isoproxil fumarate 300 mg and another NRTI in this combination are effective against hepatitis B; thus, withdrawal of the combination in those patients infected with hepatitis B may worsen the condition.
How Vonaday Tablet Works
Efavirenz 600 mg tablet inhibits an enzyme known as HIV reverse transcriptase, which is required by HIV in replication of its genetic material. On the other hand, tenofovir disoproxil fumarate 300 mg tablets and lamivudine 300 mg tablets get metabolized in the body to form compounds that are analogous to nucleotides for viral DNA synthesis and prevent further replication of the viral genome.
Clinical Evidence for Vonaday Tablet
Efavirenz-based ART trials: Clinical trials in large patients demonstrated that the efavirenz 600mg tablet with the two NRTIs was able to reduce the viral load of HIV. This made the efavirenz 600 mg-based treatment a valuable first-line treatment strategy previously.
Modern comparative trials: However, more recent research indicated that dolutegravir-based regimens have better resistance and tolerability profiles.
Side Effects of Vonaday Tablet
Vonaday Tablet side effects may be varied, depending on the dosage and patient reaction to the medication. Side effects can be managed, but some may need immediate medical attention. These include:
Common side effects
- Dizziness
- Vivid dreams or sleep disturbance
- Headache
- Nausea

- Difficulty concentrating
Serious side effects
- Severe psychiatric symptoms
- Severe skin reactions
- Hepatotoxicity

Critical Safety Warning
CNS/PSYCHIATRIC AND HBV WARNING: Efavirenz 600mg tablets may lead to notable side effects in the brain and nervous system, particularly at the start of treatment. Tenofovir disoproxil fumarate 300 mg-based treatment must not be discontinued abruptly in patients having hepatitis B, since it leads to an increase in the condition, thereby causing serious liver complications.
Vonaday Tablet Dosage and How To Take It
Important: Dose and regimen must be prescribed by the treating specialist. Tablet/vial strength is not necessarily the patient dose. Do not start, stop, split, combine or substitute treatment based on website information.
| Clinical situation | Dose |
|---|---|
| Fixed dose | One tablet once daily when this complete regimen is clinically appropriate. |
| Administration | Often taken on an empty stomach at bedtime to reduce efavirenz-related CNS adverse effects. |
| Renal impairment | Fixed-dose combination may be unsuitable when component dose adjustment is needed. |
| Switching | Do not switch from a suppressive modern regimen solely for convenience without resistance/history review. |
Vonaday Tablet Missed Dose
Take a missed dose when remembered unless close to the next dose. Do not double. Maintaining daily adherence is critical to avoid viral rebound and resistance.
Safety Advice for Vonaday Tablet
| Category | Status | Guidance |
|---|---|---|
| Pregnancy | Specialist guided | Vonaday Table has already been evaluated during pregnancy, but the physician should select the regimen according to the recommendations for HIV therapy. |
| Kidney | Monitor | As Tenofovir disoproxil fumarate 300 mg tablet may damage the kidney, a renal function test must be done when taking the medicine. |
| Liver | Monitor | Efavirenz tablet can cause liver problems, so liver tests may be needed. |
| Mental health | Caution | Report severe depression, suicidal thoughts, hallucinations or confusion to the doctor immediately. |
| Driving | Avoid if affected | Dizziness and difficulty concentrating can occur, especially early in treatment, so avoid driving if these effects occur. |
Monitoring During Vonaday Tablet Treatment
- HIV viral load will help evaluate how the therapy is managing the infection.
- The CD4 count test may be performed when necessary to assess the status of the immune system.
- The kidney function must be evaluated because the tenofovir disoproxil fumarate tablet has an impact on the kidneys.
- The status of the liver needs to be assessed because the efavirenz 600mg tablet may lead to liver dysfunction.
- The hepatitis B virus (HBV) infection needs to be controlled, particularly in patients with HIV/HBV co-infection.
- Psychological and nervous system status needs to be evaluated, including mood disturbances, dizziness, confusion, and sleep disorders.

FAQs About Vonaday Tablet
Can Vonaday Tablet be used as a full HIV treatment program?
Yes, as long as its doses are appropriate and there is no resistance or contraindication to its use.
Why is the Lamivudine 300 mg tablet commonly administered at night?
The drug efavirenz 600mg tablet may lead to dizziness, insomnia, and vivid dreams, particularly in the early stage of the medication use. However, its night intake may help deal with such side effects more easily.
Can Tenofovir disoproxil fumarate 300mg tablet still be considered an optimal treatment for HIV?
Most current HIV guidelines recommend treatment based on dolutegravir, but efavirenz 600mg-based treatment regimens can still be considered as an option in some cases.
Why is it important to know about the hepatitis B status with Vonaday Tablet?
Tenofovir disoproxil fumarate tablet and NRTI drugs included in the medication also inhibit hepatitis B, and its sudden stoppage leads to hepatitis B reactivation.
Can the use of Vonaday Tablet be discontinued if there is no viral load detected?
No. Viral load may be controlled using the medication but not eradicated. It is possible that stopping treatment might lead to an increase in the viral load once more.
How to deal with a missed dose?
It can be taken as soon as it is realized but if it is close to the time for taking the next dose, then it should be skipped and not double up the doses.
Does Vonaday Tablet have any interaction with other medicines?
Yes. Efavirenz 600mg is known to interact with some medications by altering the way they are metabolized in the body.
