Overview

Lenangio 5mg Capsule contains lenalidomide 5 mg, an immunomodulatory anticancer medicine derived from thalidomide. It binds cereblon and changes the degradation of transcription factors involved in malignant plasma/B-cell survival and immune signaling. It is widely used in multiple myeloma and selected myelodysplastic syndromes and lymphomas. Lenalidomide 5mg capsules can cause severe embryo-fetal toxicity, neutropenia, thrombocytopenia, and venous/arterial thrombosis. The 5 mg capsule is a strength; dose and cycle depend strongly on indication and kidney function.

Uses and Indications

In multiple myeloma, the lenalidomide 5mg capsule is used with dexamethasone and other agents in newly diagnosed or relapsed disease and as maintenance after autologous stem-cell transplantation. In transfusion-dependent anemia due to low/intermediate-1-risk MDS with deletion 5q, 5 mg is a standard starting dose under the label. It is also used with rituximab for follicular lymphoma and marginal-zone lymphoma in selected settings.

How Lenangio 5mg Capsule Works

Lenalidomide 5mg Capsule binds cereblon, altering the ubiquitin ligase complex and promoting degradation of IKZF1/IKZF3. This has direct antimyeloma effects and enhances T-cell/NK-cell immune activity.

Clinical Evidence for Lenangio 5mg Capsule

FIRST/MM-020: Lenalidomide-dexamethasone established an effective backbone for transplant-ineligible newly diagnosed myeloma.

CALGB 100104/IFM maintenance studies: Lenalidomide 5mg capsule maintenance after autologous transplant prolonged progression-free survival.

MDS del(5q) studies: Lenalidomide 5mg Capsule produced high rates of transfusion independence and cytogenetic response in selected del(5q) MDS.

Side Effects of Lenangio 5mg Capsule

Just like any other medication, the Lenangio 5mg Capsule is also associated with some side effects. Most of the side effects are minor and usually go away within some time. Here are some of the symptoms, including:

Common side effects

  • Neutropenia
  • Thrombocytopenia
  • Anemia
  • Fatigue
  • Diarrhea/constipation
  • Rash
  • Muscle cramps

Serious side effects

  • Severe embryo-fetal toxicity
  • Venous/arterial thrombosis
  • Serious infection
  • Severe skin reactions
  • Tumor lysis/tumour flare in lymphoma
  • Secondary primary malignancies

Critical Safety Warning

TERATOGENICITY AND THROMBOSIS: lenalidomide 5mg Capsule is structurally related to thalidomide and can cause severe fetal harm. Strict pregnancy-prevention controls are required. Myeloma combinations with dexamethasone increase clot risk and commonly require thromboprophylaxis based on individual risk.

Lenangio 5mg Capsule Dosage and How To Take It

Important: Dose and regimen must be prescribed by the treating specialist. Capsule/vial strength is not necessarily the patient dose. Do not start, stop, split, combine, or substitute treatment based on website information.

Clinical situationDose
Myeloma common regimen25 mg once daily on Days 1–21 of a 28-day cycle with dexamethasone in many regimens; modern combinations vary.
Post-transplant maintenanceCommonly 10 mg once daily continuously, with possible escalation if tolerated.
MDS del(5q)10 mg once daily; schedule/dose may be modified for cytopenias.
Renal impairmentDose reduction is required because the lenalidomide is renally eliminated.

Lenangio 5mg Capsule Missed Dose

Follow the specific regimen for missed doses; do not double. Because some labels use a time threshold for taking a late dose, follow the pack leaflet or contact the hematology team.
